Hammond was born at Chertsey in Surrey on 18 August 1605, the youngest son of John Hammond (c. 1555–1617), physician to the royal household under King James I.
His brother was Judge Thomas Hammond, a regicide of King Charles I. He was educated at Eton College, and from age 13 at Magdalen College, Oxford, becoming demy or scholar in 1619. On 11 December 1622 he graduated B.A. (M.A. 30 June 1625, B.D. 28 January 1634, and D.D. in March 1639), and in 1625 was elected a fellow of the college.
He took Holy Orders in 1629.
Hammond was a pioneer Anglican theologian.
He was the first English scholar to compare manuscripts of the New Testament.
